amyotrophic lateral sclerosis (ALS)

We are looking for you to join us full-time for a position that is initiÂally limited to two years, with the option for extension.
The position will be based at the DZNE Ulm site, which studies the underÂlying mechanisms of disease causation with a partiÂcular focus on amyoÂtrophic lateral sclerosis (ALS) and Parkinson’s disease. Using next-generation sequenÂcing, we have identified Mendelian disease genes that cause autoÂsomal-dominantly inÂherited ALS. We have developed innoÂvative disease models based on these findings (e.g. Freischmidt et al., Nat Neurosci, 2015, Brenner et al., Brain, 2018; Brenner et al., J Exp Med, 2024). We will bring these analyses to the next level by studyÂing the contriÂbution of somatic (mosaic) mutaÂtions that may occur during developÂment or aging to ALS. To this end, we are offeÂring a PhD position to employ tarÂgeÂted deep sequencing or single cell-resolved detecÂtion of mosaic mutations with low allele freÂquency in human post-mortem tissue to gain further insights into the role of genetic mosaÂics in neuroÂdegeneration. Another aim of this proÂject will be the developÂment of in vitro-models for somatic mutations for functiÂonal studies or the developÂment of diagnostic approaches for ante-mortem detection of, and potenÂtial testing of theraÂpeutic interÂventions against, somatic mutations in neuroÂdegenerative diseases such as ALS.
Your role in detail
- Perform and analyze deep sequenÂcing experiments on autoÂpsy material from ALS patients
- Make use of our established spatial transcripÂtomic expertise to study mosaic mutaÂtions and their downÂstream consequences in brain tissue
- Learn and employ iPSC-based techniques
- Perform functional experiÂments on genetic mosaics and test theraÂpeutic approaches in vitro
- Development of user-friendly tools for data analysis and tracking of experiments
- Prepare and write manusÂcripts for publication in interÂnational scientific journals
- Oral presentation of results on local, national and interÂnational conferences
- Interface between scientists working in the lab and bioÂinformatical analysis
- Master’s degree in biology, moleÂcular medicine or a related field
- Technical knowledge on the field of cell bioÂlogy, genoÂmics and sequenÂcing and its impliÂcations on data analysis would ideal
- Strong willingness to learn new tools for data analysis
- A keen interest in neuroÂdegenerative diseases
- English language is a prerequisite
The successful candiÂdate will be assisting to coordinate and direct the project. ThereÂfore, excellent communiÂcation skills and team worÂking skills are mandatory.
